AOD-9604 PEPTIDE · THE ESSENTIALS

What AOD-9604 is
and what it might help with.

It has mainly been studied for fat metabolism. The idea is to influence the use of stored fat without reproducing every effect of growth hormone. A human weight-loss benefit has not been confirmed.

A modified fragment studied in its own right

AOD-9604 is a synthetic peptide containing 16 amino acids. It is based on the end of human growth hormone — amino acids 177–191 — with an added tyrosine. The full hormone contains 191 amino acids. Such a small fragment does not automatically inherit all its functions. ↗

Sources may call it AOD9604 or LAT8881; the latter name appears mainly in pain research. AOD-9401 is a different fragment, while a commercial “HGH Fragment 176–191” label alone does not establish a product’s identity or quality. ↗

Body fat

There are favourable experimental findings. The larger clinical trial did not confirm weight-loss efficacy. ↗

Blood sugar

Researchers examined differences from full growth hormone. A diabetes treatment has not been established. ↗

Cartilage and pain

These are separate research directions. Animal findings and clinical results need separate interpretation. ↗

MECHANISM · SIMPLY EXPLAINED

How it might affect
fat metabolism.

Adipose tissue stores energy as fat. Researchers examined two processes: releasing stored fat and building new reserves.

Explanatory schematic. It is not an injection route, an established receptor map or a weight-loss simulation.

What lipolysis means

Lipolysis is the breakdown of fat stored in cells. Released fatty acids can be used as fuel. Experiments linked AOD-9604 to increased breakdown and reduced formation of new fat. ↗

What we know about receptors

Mouse research implicated the beta-3 adrenergic pathway in some metabolic effects. This does not prove direct binding to that receptor in humans. It does not have semaglutide’s GLP-1 mechanism or tirzepatide’s dual mechanism. ↗

Why a promising mechanism is only a starting point

The body can use, store again or compensate for released energy. Changes in a cell therefore cannot predict how many kilograms someone will lose. A practical answer requires comparison with placebo over an adequate period. ↗

WEIGHT LOSS · FROM ANIMALS TO PEOPLE

What the studies show
about weight loss.

Early studies supported further investigation. Testing in a larger group of people did not confirm the hoped-for result.

01 / ANIMALS

Less weight gain

In the 2000 Ng study, obese rats receiving the peptide gained less weight over 19 days. The animals still gained weight. This does not mean they lost weight or that humans would lose a corresponding percentage of fat. ↗

02 / HUMANS · METAOD005
300participants · 12 weeks

An early clinical signal

This oral study compared several doses with placebo. A conference abstract reported favourable results in some groups. Incomplete publication limits how confidently that signal can be interpreted. ↗

03 / HUMANS · OPTIONS / METAOD006
502randomised participants · 24 weeks

Weight-loss benefit was not confirmed

Oral AOD-9604 was compared with placebo; all groups received a diet and exercise programme. The primary assessment was at 12 weeks, with follow-up at 24. The peptide did not demonstrate significantly greater weight loss. The obesity programme was discontinued in 2007. ↗

Does it help with belly fat?

There is no robust clinical demonstration of selective abdominal fat loss or an added benefit on top of GLP-1 treatment. Accounts of a smaller waist need to be read alongside diet, exercise and concurrent substances. ↗

This comparison shows stages of research. Duration and participant numbers describe the studies; they do not indicate the size of any weight effect.

GLUCOSE · IGF-1 · GROWTH HORMONE

How it differs
from growth hormone.

AOD-9604 was designed to explore a narrower set of metabolic effects. Its origin does not make it a replacement for GH.

What researchers measured

Historical trials monitored blood glucose, glucose tolerance and IGF-1. The safety paper did not find a pattern of raised IGF-1 or impaired glucose tolerance under the studied conditions. Episodes of hypoglycaemia were nevertheless reported; saying it never affects glucose would be too strong. ↗

What this means for you

These findings do not establish muscle growth, treatment of hormone deficiency or prevention of diabetes. Nor do they establish the safety of combinations with insulin or other medicines. The pancreas is an anatomical reference for glucose regulation, not an established AOD-9604 treatment target. ↗

JOINTS · ANIMAL RESEARCH

Why the peptide also appears
in cartilage research.

A 2015 study included 32 rabbits with experimentally induced osteoarthritis, divided into four groups.

AOD-9604 alone and with hyaluronic acid

Researchers compared saline, hyaluronic acid, AOD-9604 and the combination, with eight animals in each group. They assessed cartilage and limping. The combination produced the most favourable reported findings. ↗

Administration was into the joint, in an animal model. The result cannot be transferred directly to a subcutaneous injection and does not establish efficacy in human osteoarthritis. It supports research interest, not a promise of knee regeneration. ↗

LAT8881 · THE SAME MOLECULE

What was investigated
for pain.

Under the name LAT8881, the peptide was also tested for neuropathic pain — pain linked to nerve damage. This is a different question from weight loss.

ORAL TRIAL · 53 ADULTS

Mean change in pain after four weeks

This result does not establish a clinical benefit for neuropathic pain. It also provides no estimate of weight loss. Other LAT8881 indications require their own evidence. ↗

PERSONAL EXPERIENCES · ONLINE ACCOUNTS

What people say
after trying it.

Accounts include favourable experiences, no noticeable benefit and discomfort. Many people were already using other peptides. These are paraphrases of public accounts, not quotations or a representative survey.

01 / Reddit

A waist change alongside tirzepatide

The author was already using tirzepatide for weight maintenance and wanted a change around the waist. After nearly four weeks with AOD-9604, they described a change they welcomed. With overlapping treatments, the account cannot show each one’s contribution.

Seven weeks without an added effect

The author was using Ozempic and had reached a weight plateau. They added AOD-9604 hoping weight loss would resume, but reported no difference after seven weeks. They asked others what a noticeable benefit should actually look or feel like.

Local pain after the first injection

One person described pain almost immediately at and around an injection site, reportedly lasting several hours. Replies described different experiences. The preparation was not independently analysed, so the peptide, formulation and other factors cannot be disentangled.

Product composition and authors’ measurements are unverified. These accounts cannot establish the chance of weight loss or the frequency of adverse reactions.

SAFETY · AVAILABLE EVIDENCE

Safety and possible
adverse effects.

Some information comes from historical oral or intravenous trials. Those findings do not automatically establish the safety of currently marketed subcutaneous preparations.

What symptoms were reported

Trials reported headache, digestive symptoms, fatigue and dizziness. Some published percentages pool every group, including placebo. They are not the proportion experiencing an AOD-9604-caused reaction. Online accounts describe injection pain, without a validated clinical frequency. ↗

What the serious-event data show

METAOD005 also recorded serious events, including tumour diagnoses. Investigators considered them unrelated to treatment; FDA judged the information insufficient to independently exclude a relationship. This is uncertainty, not proof that the peptide causes cancer. ↗

The preparation matters too

Purity, aggregation and degradation can change a peptide’s properties and potential immune reactions. Acetate and free-base preparations are not identical in every respect. A “research use only” label or commercial certificate does not replace product-specific clinical evidence. ↗

Where important data are missing

Evidence is insufficient for pregnancy, breastfeeding, children, long-term use and many medicine combinations. These studies do not validate a self-treatment regimen. AOD-9604 is not an FDA-approved medicine; a GRAS assessment for food uses does not approve an injection. ↗

06 / FAQ

Your questions.
Clear answers.

The dossier, explained simply. Open a question for the answer and its source.

What is the AOD-9604 peptide?

AOD-9604 is a synthetic 16-amino-acid peptide derived from a modified fragment of human growth hormone. It has mainly been studied for fat metabolism. LAT8881 is another name for the same molecule.

Source & context ↗

What might AOD-9604 help with?

Research has examined fat use, body weight and, separately, cartilage and pain. Some animal experiments found favourable results, but these do not establish a human clinical benefit. The larger obesity trial did not confirm weight-loss efficacy.

Source & context ↗

Does AOD-9604 help with weight loss?

Weight-loss efficacy was not confirmed in the larger oral OPTIONS trial, with 502 randomised participants and 24 weeks of treatment. These data do not provide a number of kilograms that an individual can expect to lose.

Source & context ↗

Does AOD-9604 remove belly fat?

The sources presented do not provide robust clinical evidence of selective abdominal fat reduction. An injection site does not demonstrate a local fat-loss effect. Waist accounts can also reflect diet, exercise and other treatments.

Source & context ↗

Is AOD-9604 the same as growth hormone?

No. Full growth hormone contains 191 amino acids; AOD-9604 contains 16. The peptide is not an established GH replacement or a secretagogue — a substance used to stimulate growth-hormone release.

Source & context ↗

Does it increase muscle mass or IGF-1?

The evidence presented does not establish muscle growth in humans. Safety studies did not identify a pattern of raised IGF-1 under the tested conditions. Its growth-hormone origin does not justify promises of anabolic effects.

Source & context ↗

Does AOD-9604 affect blood glucose?

Historical trials did not show a pattern of impaired glucose tolerance, but episodes of hypoglycaemia were reported. It is too strong to say it cannot affect glucose at all. Efficacy as a diabetes treatment has not been established.

Source & context ↗

How does it differ from semaglutide or tirzepatide?

AOD-9604 is a GH-derived fragment investigated for fat metabolism. Semaglutide acts on GLP-1; tirzepatide acts on GIP and GLP-1. No direct trial presented establishes equivalence or an added benefit on top of those medicines.

Source & context ↗

Can it repair damaged cartilage?

A study in 32 rabbits with induced osteoarthritis reported favourable findings, especially with hyaluronic acid. Administration was into the joint. This is not a human clinical result and does not validate subcutaneous injection for cartilage repair.

Source & context ↗

Why is it also called LAT8881?

LAT8881 is an AOD-9604 synonym used in pain research. An oral crossover study in 53 adults did not find a significant advantage over placebo for neuropathic pain. Pain research does not establish a weight effect.

Source & context ↗

What adverse effects have been reported?

Trials reported headache, digestive symptoms, fatigue and dizziness. Some serious events remained insufficiently clarified. Clinical data for subcutaneous products are inadequate; online reports of local pain do not provide a validated frequency.

Source & context ↗

Is AOD-9604 FDA approved? What about GRAS?

AOD-9604 is not an FDA-approved medicine in the sources checked for this dossier. A GRAS assessment concerns specific food uses; it is not approval of an injectable medicine or evidence of weight-loss efficacy.
