MELANOTAN II · THE ESSENTIALS

Skin pigmentation and interest in a tanned appearance

Melanotan II is an experimental peptide that can stimulate skin pigmentation. Interest often begins with a very human wish: a deeper colour and greater confidence in one's appearance. On forums, some people greatly appreciate the change; others find that their colour, freckles or how they feel develop differently from what they imagined. ↗ ↗ ↗

There is early human research on pigmentation and sexual responses, but the clinical evidence base is small. Understanding the signal the molecule sends and the outcomes actually measured helps put photographs and personal experiences into context.

A small peptide with a distinct structure.

Melanotan II, also called MT-II or MT2, is a synthetic analogue of α-MSH, a hormone involved in melanocortin signalling. It has seven amino acids and a structure partly closed into a ring. This identity distinguishes it from other molecules with similar names. ↗

Seven amino-acid residues, six of which form the ring. The Asp–Lys bridge closes the cyclic portion. Nle means norleucine, while D-Phe identifies that residue’s form. ↗

MECHANISM · EXPLAINED SIMPLY

From signal to pigment.

The molecule influences melanocortin receptors, through which cells receive signals from this hormone family. In skin, MC1R is linked to the activity of melanocytes, the cells that produce pigment. MT-II's activity at multiple receptors helps explain effects beyond skin colour. ↗

Conceptual diagram. Dots and distances do not represent quantities, speed or tanning intensity.

A family of receptors.

MT-II influences melanocortin receptors. MC1R is relevant to skin pigmentation, while activity at several receptors helps explain effects beyond colour. This diagram shows biological relationships without simulating the strength of a personal response. ↗

The cells that produce pigment.

Melanocytes produce melanin, the skin pigment. The signal can stimulate the system producing colour; the human pilot observed increased pigmentation in some participants. The circles in the diagram represent pigment conceptually, without clinical values, timing or proportions. ↗

Appearance and tolerability belong together.

A visible change may be appreciated, but distribution and tolerability can vary. Experiences include greater confidence in appearance, more noticeable freckles or nausea. Small studies and selected accounts cannot predict an individual outcome. ↗ ↗

Human studies observed pigmentation, erectile responses and effects such as nausea, yawning or reduced appetite. They did not directly measure every pathway in the brain. An explanation of the mechanism needs to preserve this distinction between signalling studied in the laboratory and effects observed clinically. ↗ ↗ ↗

PIGMENTATION · HUMAN RESEARCH

An early clinical signal.

The first study is small but important for understanding where the interest begins. It observed skin colour under research conditions, with saline as control.

VOLUNTEERS · PHASE I · 1996

Human pigmentation. In the phase I pilot published by Dorr and colleagues, three healthy men received MT-II and saline on alternating days. Increased pigmentation was observed in two participants. This is a clinical starting point for the aesthetic interest; the sample cannot provide a credible success rate or predict the shade obtained. ↗

A numerical reflectance series was not extracted from the article. The result is therefore explained in text, without an invented tanning curve or success percentage.

Phase I, single-blind, three healthy volunteers; subcutaneous MT-II versus saline on alternating days for two weeks. Pigmentation was assessed using reflectance and visual observation one week after completion. Funding: NCI/NIH, CA27502. Original abstract and metadata consulted; full text was inaccessible. An interesting laboratory direction.

An interesting laboratory direction. Wu and colleagues observed slower growth of melanoma tumours in mice following topical MT-II application. The finding explores a cellular pathway and deserves mention. It did not test cancer prevention and does not establish a human treatment. The article also contains an inconsistency about the experiment's duration, so its tumour values are not proposed here as a time-based chart. ↗

How does this fit with melanoma reports? Clinical cases have been temporally associated with use, sometimes alongside sunbeds. They warrant attention but do not, by themselves, prove that MT-II caused cancer. The mouse experiment and these cases address different questions; neither establishes the safety of recreational use. ↗ ↗ ↗

Wu et al. · 2020 · cells and mice

Topical MT-II versus PBS, six mice per group, established B16-F10 tumours. In vitro cell proliferation was not reduced, although other tumour behaviours were affected. APPEARANCE IN EVERYDAY LIFE

Shade, freckles and how you see yourself.

An appreciated shade can come with differences between areas. More visible freckles are a welcome change for some people and an unwanted outcome for others. The actual appearance matters, beyond simply having darker skin. ↗ ↗

After stopping

After stopping, the duration and extent of fading are not well established. Accounts of darker areas or persistent freckles do not demonstrate that the molecule keeps acting for months. ↗ ↗

Colour and UV exposure

Colour does not replace sun protection. Use alongside sun or sunbeds complicates interpretation and does not make UV exposure safe. A changing mole deserves dermatological assessment. ↗ ↗

SEXUAL RESPONSE · SMALL HUMAN STUDIES

Another effect, measured separately.

These studies examined erectile response in men with erectile dysfunction. The charts describe that outcome, not skin colour, sun protection or a healthy person’s performance.

An acute erectile response. In the 1998 study, involving ten men with erectile dysfunction without a known organic cause, mean time with penile tip rigidity above 80% was 38.0 minutes with MT-II and 3.0 minutes with placebo during six hours of observation. The 80% threshold describes measured rigidity, not the proportion of people who responded. ↗

Another clinical group. In the 2000 study, ten men with organic risk factors had 45.3 minutes at the same rigidity threshold, compared with 1.9 minutes with placebo. Subjectively reported erections occurred after 12 of 19 active injections and 1 of 21 placebo injections. The denominators are repeated injections in the same participants. This study is presented separately from the 1998 trial. ↗

MOLECULES & STATUS

Similar names, distinct molecules.

Scenesse contains afamelanotide, a different molecule approved in the EU to prevent phototoxicity in adults with erythropoietic protoporphyria, a rare disease causing painful reactions to light. Vyleesi contains bremelanotide, also known as PT-141, approved in the US for a specific sexual desire disorder in premenopausal women. These approvals do not apply to Melanotan II. ↗ ↗

The official sources consulted do not identify an approved indication for MT-II in the EU or US. The European warning consulted is older; the status is presented within the limits of available sources, without claiming an exhaustive check of every national register. The FDA continues to describe safety concerns in the compounding context. Pharmacy availability and reimbursement in Romania have not been confirmed. ↗ ↗ ↗

Checked: 15 October 2026. Approval, availability and reimbursement require separate checks.

PERSONAL EXPERIENCES · REDDIT & BLUELIGHT

What people appreciate and what surprises them.

Five experiences actually read and paraphrased. These are selected perspectives, without verification of products, diagnoses or colour under standardised conditions. They do not show how often an outcome occurs.

01 / Reddit

greater confidence in appearance

A person describing themselves as very pale, with freckles, appreciates the deeper colour and says they felt more comfortable without the makeup used to cover their paleness. In the same thread, they explain that the result was uneven and their abdomen had become too dark. Enjoyment of the change and its aesthetic limitation belong to the same experience.

02 / Bluelight

a colour appreciated over time

A person describes a tan they consider natural and few additional freckles during a period of continued use. The context includes sunbeds and, in the earlier account, other substances. Appreciation of the colour is clear, but the experience does not separate each exposure's contribution or demonstrate protection against burning or long-term safety.

more colour than intended

A person wanted a subtle change but noticed freckles and differences in colour between areas. Their family noticed a stronger tan than they perceived themselves, and use was stopped. Later, they felt their neck had remained much darker. This observation does not prove that the molecule continued acting for months.

04 / Bluelight

appearance appreciated, reactions difficult to tolerate

A person who says they usually tanned poorly appreciated the colour obtained but stopped because of nausea, perceived changes in pulse and blood pressure, and more noticeable facial freckles. In a later follow-up, the freckles had faded but remained visible. They specify that they had not developed new moles; freckles and moles remain distinct observations.

a good experience initially, then stopping

A person who already tanned and used sunbeds says they initially liked the deeper shade. Later, they describe hives, flushing, nausea and a racing heart, followed by stopping use. Their subsequent fear that they could no longer tan naturally has not been medically confirmed. The product's composition and the cause of the symptoms were not verified.

SAFETY

Safety and possible adverse effects.

Nausea, yawning, stretching and reduced appetite appear in early human research; the pilot also described sleepiness or fatigue. In the 2000 study, severe nausea was associated with 4 of 19 active injections. The abstract does not provide the corresponding placebo count. This figure is not a percentage of users and cannot be extrapolated to current products. ↗ ↗ ↗

Safety reports include pigmentation changes, priapism and serious neurological or renal events. Cases do not provide a reliable frequency or automatically establish causation. The FDA also highlights peptide impurities or aggregation, which may promote immune reactions. ↗ ↗ ↗

Product quality is a concrete uncertainty. The TGA tested five nasal bottles labelled 30 mg and found estimated content ranging from 22 to 54 mg, based on the labelled volume. The finding concerns those bottles; it does not measure the amount absorbed or describe all products. ↗

Estimated range: 22–54 mgLabel: 30 mg

Estimates based on the labelled 20 mL volume. Individual values for the 5 bottles were not published. The range does not measure absorption or efficacy and does not describe all products. ↗

We do not have an approved MT-II label from which to extract validated contraindications and interactions. The selected data do not establish safety during pregnancy, breastfeeding or childhood, or provide a verified contraception interval. Rules for Scenesse or Vyleesi do not automatically transfer. Existing illnesses, concurrent medicines and changing moles need discussion with a clinician. ↗ ↗ ↗

An erection persisting towards 3–4 hours requires emergency assessment, particularly if painful. Difficulty breathing, tongue or throat swelling, fainting or sudden neurological symptoms require urgent help: in Romania, call 112 or attend an emergency department. These are signs for recognising emergencies, without automatically attributing their cause to MT-II. ↗ ↗ ↗

06 / FAQ

Your questions.
Clear answers.

The dossier, explained simply. Open a question for the answer and its source.

Can Melanotan II deepen skin colour?

Yes, there is an early human signal of increased pigmentation. The pilot study observed a change in some participants, and online experiences sometimes describe appreciated results. However, the available research cannot support promises of a particular shade, evenness or success rate. The appearance someone hopes for and the result they actually experience may differ.

Source & context ↗

How soon might a change be visible?

There is no predictable timeline for every person. The pilot reported increased pigmentation one week after a two-week study period ended. This is the assessment time in that experiment, not a promise that effects begin on a particular day. Products, UV exposure and individual responses complicate any comparison with online accounts.

Source & context ↗

What happens to the colour after stopping?

The duration and extent of fading are not well established in the selected human studies. Accounts describe different developments, including freckles remaining visible or areas perceived as darker. Without standardised measurements and a history of sun exposure, these observations demonstrate neither permanent pigmentation nor that the substance remains active for months.

Source & context ↗

Is it the same as Melanotan I or Scenesse?

No, Scenesse contains afamelanotide, a distinct molecule historically associated with the name Melanotan I. In the EU, the implant is approved to prevent phototoxicity in adults with erythropoietic protoporphyria. This medical indication is not an approval of MT-II for tanning. The implant's results and rules for use do not automatically transfer between molecules either.

Source & context ↗

Is it the same peptide as PT-141?

No, PT-141 is bremelanotide, the molecule in Vyleesi. The FDA approved that medicine for a specific population of premenopausal women with a particular sexual desire disorder. Their relationship to the melanocortin family explains some similarities, but this association does not give MT-II Vyleesi's approval, demonstrated benefits or adverse-effect profile.

Source & context ↗

Does it work for very pale people who tan poorly?

There are insufficient clinical data to predict this group's response. One account read describes a change appreciated by a very pale person, alongside uneven pigmentation. Their skin type was not clinically confirmed. The small pilot provides no robust analysis by skin type, so one favourable experience cannot become a rule for all pale people.

Source & context ↗

Can it make freckles or moles more noticeable?

Yes, more noticeable pigmented areas are described in reports and personal experiences. More visible freckles are not automatically new moles, and a colour change is not a cancer diagnosis. A lesion changing shape, size or colour needs dermatological assessment. Online accounts cannot establish the nature of a lesion or the cause of its change.

Source & context ↗

Does it replace sun protection or make sunbeds safer?

No, the colour obtained does not replace sun protection or demonstrate sunbed safety. Some experiences include UV exposure, making it difficult to separate the peptide's effect from light's contribution. The selected human studies did not demonstrate skin cancer prevention through recreational use. A more tanned appearance does not measure the skin's actual protection.

Source & context ↗

Does nasal spray have the same evidence as the studied injections?

No, results from subcutaneous studies do not automatically transfer to a nasal spray. Formulation, absorption and actual content are separate questions. TGA tests identified content variation in nasal products without measuring efficacy. An account read also reported little or no visible effect; it does not prove that all sprays work or that all are ineffective.

Source & context ↗

Does reduced appetite mean it helps with weight loss?

No, reduced appetite observed in early research does not demonstrate sustained weight loss. The selected studies examined other outcomes and provide no validated clinical result for treating obesity. Nausea may accompany appetite changes and limit tolerability. Eating less for a short period is not the same as a demonstrated metabolic benefit.

Source & context ↗

Are the sexual effects real?

Yes, an acute erectile response was measured in two small studies involving men with erectile dysfunction. These findings do not establish lasting benefits in healthy people or constitute approval for improving sexual performance. Erection and desire remain different outcomes. A persistent or painful erection may require urgent medical attention.

Source & context ↗

Does it cause or prevent melanoma?

Neither claim is demonstrated by the data consulted. There are melanoma reports following use, sometimes alongside sunbed exposure, without sufficient evidence of causation. Separately, a topical experiment in mice slowed the growth of existing tumours. It did not test cancer prevention and does not establish safety or treatment in humans.
